FAQ

1. ACRO的细胞株产品是单克隆还是细胞池(pool)？

我们的细胞株产品均为单克隆细胞株。

2. 细胞株的运输条件是什么？到货后细胞株储存和使用应该注意什么？

细胞株通过干冰运输。为了确保细胞的最佳活力，我们建议您收到细胞后立即解冻并开始培养。若无法立即解冻培养，建议您立即转移到液氮中长期储存，请注意转移过程要迅速，不要造成解冻，避免影响细胞长期的稳定性和活力。如果不能马上转移到液氮中，可暂时存放于-80°C冷冻冰箱，我们建议最长保存时间自到货之日算起不超过2周，不建议长期保存于干冰或-80°C冷冻冰箱中。使用细胞时，请参照DS中推荐的复苏培养方法，进行细胞的复苏。

请注意: 如果收到时细胞未冷冻或未置于干冰中，请及时联系技术支持 tech.cn@acrobiosystems.com 。

3. 购买了一款细胞株产品，为什么收到的是两支细胞冻存管？

我们提供两支细胞冻存管是为了确保您的实验顺利进行。万一第一支冻存管的细胞解冻复苏培养出现异常情况，请联系技术支持（tech.cn@acrobiosystems.com）进行问题排查后再解冻第二支。所有ACRO细胞株在发货前均已做过复苏培养验证，同时我们建议在尽可能早的传代阶段建立细胞库，以便长期使用。

4. 收到细胞株后，进行细胞的复苏需要注意什么？

细胞解冻后，首先应在不含筛选抗生素的培养基中维持培养1-2代，如果细胞状态良好，后续可使用添加筛选抗生素的培养基进行传代培养。对于筛选抗生素的选择请参考FAQ11。建议在细胞培养的整个过程中可添加双抗P/S (Penicillin-Streptomycin)。

5. 细胞的传代需要注意什么？

- 贴壁细胞，主要针对HEK293细胞：培养汇合度不宜过高，如汇合度过高（超过100%），可能会严重影响传代后的细胞状态，具体操作请参考DS中的传代方法及注意事项。如果因汇合度过高或其他原因导致传代后细胞贴壁较差，建议在培养基中去掉筛选抗生素，同时按照较高的细胞量，建议T75培养瓶按1×107个的细胞总量进行传代培养，待细胞状态恢复正常后再进行正常的传代培养。

- 悬浮细胞，主要针对Jurkat和Raji细胞：细胞密度不宜过高，如密度过高（超过3×106 cells/mL），可能会严重影响传代后的细胞状态，具体操作请参考DS中的传代方法及注意事项。如果因细胞密度过高或其他原因导致传代后细胞活率较差，建议在培养基中去掉筛选抗生素，同时按照较低的密度，建议按1×105-2×105/mL进行传代培养，待细胞状态恢复正常后再进行正常的传代培养。

6. 初期进行功能实验时建议使用哪种培养板进行贴壁细胞的种植？

我们建议首先选择透明的96孔板进行细胞种植（具体参照实验Protocol），以便观察细胞状态并判断细胞量是否合适。待实验条件摸索成熟后，可尝试其他适合的培养板。种植板细胞量的推荐请参考FAQ8。部分常用培养板等耗材，详见产品实验protocol。

7. 进行功能实验时，细胞种植板时培养基是否还需要添加筛选抗生素？

无需添加筛选抗生素。

8. 对于贴壁细胞，进行功能实验时按多少细胞量进行种植板比较合适？

对于96孔板，我们建议在功能实验前，让细胞过夜培养至汇合度达到约80%左右。您可以首先按照ACRO实验Protocol中推荐的细胞量进行尝试铺板或者通过设置不同细胞量的梯度，找到适合您实验系统的最优条件。

9. 功能实验中，我是否可以直接使用ACRO推荐的蛋白浓度点或者药物浓度？

在功能实验初期，可以先参照ACRO推荐的蛋白浓度或者药物浓度进行尝试摸索，但由于试剂差异或者实验条件不同，我们建议您通过前期摸索实验找到适合您实验系统的最佳浓度点。

10. 是否可以使用其它厂家的培养基和血清进行细胞培养？

我们建议优先使用DS中推荐厂家的培养基和血清。您也可以选择使用其它同类可替代的或其他合适的培养基和血清（如Gibco等品牌）尝试培养。

11. 是否可以使用其它厂家的筛选抗生素？

对于筛选抗生素，我们强烈建议选择使用DS中推荐的品牌。不同厂家抗生素产品活性可能存在差异，若使用其他品牌的筛选抗生素，需要首先验证ACRO培养方案中推荐的浓度是否合适。无论使用哪个品牌，建议同时传代一份不添加筛选抗生素的细胞进行备份，以避免因浓度不当而杀死细胞的情况。

12. 是否可以使用其它厂家的蛋白进行功能活性实验？

我们强烈建议使用实验Protocol中推荐厂家的蛋白试剂，因为我们已对其活性进行了验证。如果选择其它厂家的蛋白产品，建议参照Protocol中的推荐浓度进行浓度摸索，以找到合适的浓度点。

13. 对于细胞冻存液，你们有什么推荐吗？

对于贴壁和悬浮细胞，我们均推荐使用90% FBS+10% DMSO (V/V)作为冻存液。此外，您也可以选择商业化细胞冻存液或实验室常用的其他合适的细胞冻存液。

推荐冻存密度： 5×106 - 1×107 cells/mL
